چکیده

The increasing needs of personalized therapies have recently stimulated impressive advances in research and development of portable point-of-care biosensors for realtime monitoring as well as biosensors for drug development [1, 2]. Enzymatic amperometric biosensors based on the enzyme family cytochrome P450 (CYP) have drawn increasing attention because CYP is a monooxygenase enzyme primarily involved in the metabolism of bioactive metabolites and hydrophobic xenobiotics such as drugs, environmental pollutants and steroids [3]. The 3D structure of the enzyme is illustrated in Scheme 1A. CYP catalyses the monooxygenase reaction, a two-electron reaction where the electrocatalytic transformation of a substrate is coupled to the electrocatalytic reduction of oxygen, according to the general equation:

Multi-panel drugs detection in human serum for personalized therapy.

This work focuses on P450 biosensors based on multiwalled carbon nanotubes (MWCNT) and different cytochrome isoforms: 3A4, 2B4, 2C9. The proposed biosensors exhibit enhanced sensitivities and decreased detection limits thanks to carbon nanotubes. The MWCNT structuring improves the sensitivity from 5.1 to 20.5 nA/mM mm(2) in case of CYP2B4-mediated Benzphetamine detection, from 0.26 to 0.63 nA/μ...

Electrochemical Detection of Anti-Breast-Cancer Agents in Human Serum by Cytochrome P450-Coated Carbon Nanotubes

We report on the electrochemical detection of anti-cancer drugs in human serum with sensitivity values in the range of 8-925 nA/μM. Multi-walled carbon nanotubes were functionalized with three different cytochrome P450 isoforms (CYP1A2, CYP2B6, and CYP3A4). A model used to effectively describe the cytochrome P450 deposition onto carbon nanotubes was confirmed by Monte Carlo simulations. Voltamm...

Effect of Different CNT’s Oxidation Methods on Thiocoline Detection by Surfactant Modified Graphite Electrodes

Carbon nanotubes are regarded as promising building blocks for the construction of novel biosensors due to their unique properties like fast electron transfer, high electrocatalytic effect and good biocompatibility. In the present study, the effect of different chemical oxidation’s methods of multi-walled carbon nanotubes (MWCNTs) on the electrochemical behavior of modified graphite electrodes ...

[Basic characteristics of rat brain microsomal estradiol 2-hydroxylase].

The basic properties of estradiol 2-hydroxylase in rat brain microsomes were studied and compared to the known characteristics of rat liver microsomal estradiol 2-hydroxylase. Rat liver microsomal estradiol 2-hydroxylase, which has been considered to be a cytochrome P450-like enzyme, was largely inhibited by carbon monoxide. On the other hand, the effect of carbon monoxide on the activity of ra...

Effects of gamma-hexachlorocyclohexane and L-3,3',5-triiodothyronine on rat liver cytochrome P4502E1-dependent activity and content in relation to microsomal superoxide radical generation.

Liver microsomal cytochrome P4502E1-dependent p-nitrophenol (PNP) hydroxylation and expression of cytochrome P4502E1 were studied in rats subjected to gamma-hexachlorocyclohexane (HCCH) or L-3,3,5-triiodothyronine (T3) administration as a possible mechanism contributing to superoxide radical (O2.-) generation. HCCH treatment (a single dose of 40 mg/kg body wt) produced a 43% increase in the con...
